Amylin

Also known as: islet amyloid polypeptide, IAPP, amylin peptide

Amylin, also known as islet amyloid polypeptide (IAPP), is a 37-amino-acid hormone secreted by pancreatic beta cells along with insulin. It plays a role in the regulation of energy balance following food intake and is the component of amyloid deposits found in the pancreas of individuals with type 2 diabetes.

Overview

Peptide hormone that plays a role in glycemic regulation

Amylin, or islet amyloid polypeptide (IAPP), is a 37-residue peptide hormone. It is co-secreted with insulin from the pancreatic β-cells in the ratio of approximately 100:1 (insulin:amylin). Amylin plays a role in glycemic regulation by slowing gastric emptying and promoting satiety, thereby preventing spikes in blood glucose levels after a meal.

Mechanism of Action

Amylin interacts with receptors formed by the binding of the calcitonin receptor and one of three RAMP proteins, resulting in the formation of the AMY receptor family located in the pancreas, brain, and other tissues. It is involved in glucose regulation and energy balance, influencing mechanisms such as meal-ending satiation in the brain largely via the area postrema, as well as insulin-stimulated glucose utilization in muscle tissue.

Research History

Discovered in 1986, amylin was identified as a constituent of amyloid deposits in the pancreatic islets of patients with type 2 diabetes, leading to its designation as islet amyloid polypeptide. During the 1990s, it gained recognition as a hormone co-secreted with insulin. A modified analogue named pramlintide was approved in 2005 as an adjunct to insulin therapy, prompting renewed research interest in amylin-related studies.
