Apamin

Also known as: bee venom apamin, SK channel blocker

Apamin is an 18-amino-acid peptide found in honeybee venom, comprising roughly 2 to 3 percent of the venom's dry weight. It is a highly selective blocker of small-conductance calcium-activated potassium channels, known as SK channels, and is widely used as a research tool for studying these channels in the brain, heart, and other excitable tissues.

Overview

Chemical compound

Apamin is an 18 amino acid globular peptide neurotoxin found in apitoxin (bee venom). Dry bee venom consists of 2–3% of apamin. Apamin selectively blocks SK channels, a type of Ca2+-activated K+ channel expressed in the central nervous system. Toxicity is caused by only a few amino acids, in particular cysteine1, lysine4, arginine13, arginine14 and histidine18. These amino acids are involved in the binding of apamin to the Ca2+-activated K+ channel. Due to its specificity for SK channels, apamin is used as a drug in biomedical research to study the electrical properties of SK channels and their role in the afterhyperpolarizations occurring immediately following an action potential.

Mechanism of Action

Apamin is stabilized by two disulfide bonds, which confer a compact shape that effectively targets the outer pore region of SK channels (KCa2.1, KCa2.2, and KCa2.3), thereby inhibiting potassium flow. Under normal conditions, SK channels open in response to intracellular calcium increases following cell firing, leading to potassium outflow that hyperpolarizes the cell and regulates firing frequency. By blocking this mechanism, apamin alters firing patterns, making it a valuable tool for probing SK channel function.

Research History

Apamin was isolated from bee venom in the twentieth century and became a classic pharmacological reagent after its target was identified. A 1984 review highlighted its role as a defining blocker of SK channels. Due to its unique selectivity, researchers have utilized apamin for decades to explore SK channel function, and a 2022 study revisited its structure and pharmacology using contemporary electrophysiological techniques.
