Magainin

Also known as: magainin 1, magainin 2

Magainin is a family of 23 amino acid antimicrobial peptides first found in the skin of the African clawed frog, Xenopus laevis. The two best known forms, magainin-1 and magainin-2, differ by only a few residues and are cut from the same precursor protein, which includes multiple copies of the magainin-2 sequence. UniProt catalogs the frog precursor under accession P11006, and magainin-2 has a mass near 2466.92 daltons.

Overview

Group of pore-formng peptides from the skin and intestines of frogs

The magainins are a class of antimicrobial peptides found in the African clawed frog. The peptides are cationic, generally lack a stable conformation in water but form amphipathic α-helix in membranes; their mechanism against micro-organisms is unclear but they disrupt the cell membranes of a broad spectrum of bacteria, protozoa, and fungi.

Mechanism of Action

Magainins interact with cell membranes rather than with a specific protein receptor. They gather on negatively charged membrane surfaces, insert into the lipid bilayer, and cause disruption, which restrains bacterial and fungal growth and breaks down protozoa by osmotic lysis in laboratory studies. Their membrane activity also quickly inactivates a fish herpesvirus in test systems.

Research History

Magainins emerged from studies of the frog skin defense system, the peptides the skin uses against microbes in the animal's environment. With a precursor that holds a magainin-1 section and several magainin-2 repeats, the family serves as a classic example of how amphibian skin peptides are stored in precursor form.
