FDA Approves Foundayo (Orforglipron), First Once-Daily Oral GLP-1 Pill for Weight Management
On April 1, 2026, the FDA approved Foundayo (orforglipron), an Eli Lilly once-daily pill, for weight management in adults with obesity or overweight with a weight-related medical condition. It is the first oral small-molecule GLP-1 receptor agonist to complete FDA review.
The approval breaks a long-standing pattern: until now, GLP-1 therapies were peptides requiring injection (or, in the case of oral semaglutide, strict administration conditions). Orforglipron achieves GLP-1 receptor agonism as a small molecule with no food or water restrictions.
For the research community, Foundayo’s approval is a signal about where metabolic drug development is heading. The GLP-1 pathway now has validated peptide, oral-peptide, and small-molecule formats - each with distinct research implications for receptor pharmacology and drug design.
Regulatory note: as with all FDA approvals, the decision covers the specific product and indication. It does not constitute approval of any compounded, generic, or research-grade material.
